Tuesday, June 7th: Performing splicing experiments with my RNA
Today, I am splicing with my RNA, and also digesting it with my DNAzymes, to determine how well it can digest. I am worried though that some urea got accidentally left into the sample, which could pose a problem for DNAzyme binding to the target RNA.
Monday, June 6th: Lypholizing my RNA sample
Today, I read a paper, while also lypholizing my RNA sample. This basically means to freeze dry it and sublime all the water out. I needed to concentrate it further, hence this step. I left it lypholizing overnight.
Sunday, June 5th: John and I visit the World of Coke!
John and I went on a date to the World of Coke. I enjoyed the history. Coke was invented by John Pemberton, a pharmacist, around 1886. There are pictures of Victorian ladies drinking it. At the end, was the tasting room, of all different flavors that Coke manufactures in other countries. The worst by far was the Beverly from Italy. It has a terrible bitter after taste that is absolutely vile. I have no idea who would drink the stuff.
Saturday, June 4th: preparing for our new kittens!
Today, I did laundry, cleaned the kitchen, took out the trash, ironed John's clothes - yes! Saturday is our cleaning day from the week's entropy. I also looked over a cookbook and baked cookie dough I had in the fridge.
We are going to Petsmart to pick-up supplies for the three new orange kittens we are buying from Amy Davenport. We are excited to make a home for them! We are going to get them next weekend. I was looking up what material to use in water bowls. There are so many choices! We will do our best to be good pet owners.

Friday, June 3rd: the tRNA I isolated is the correct one
Science: So, I was able to get the correct RNA, after all. It was very faint, but the band in the gel was the right size for what I expected and corresponded to the band I thought it was from the total RNA lysate.
Home: John got me frozen dumplings tonight from the Great Wall Supermarket, and I had some microwaved. They were surprisingly tasty. I've been craving these things. I wish I had proper chopsticks at home so I could eat them correctly. These weren't covered all the way in wrapping, but I liked them just the same. I ate them in soy sauce mixed with Mongolian fire oil. Very awesome.

Thursday, June 2nd: isolated a tiny amount of a tRNA
Today, I did a large EtOH precipitation and isolated a tRNA - what I hope is the one I'm interested in. However, there was hardly any of it there. Kind of discouraging. Still, I'm going to run it on a gel tomorrow to see if it's the correct one.
Nusaiba Baker officially joined the lab today! Other stories of the day: I braved the pouring rain for a cookie at Allie's Cookie Store. I decided to go home instead of staying all hours to run a gel on the tRNA.

Wednesday, June 1st: Might have got some tRNA
I'm not sure my experiment worked yet, but it looks like I isolated a little of the tRNA I hoped to isolate. It was a very small amount, but it was measurable. I went ahead and prepped it overnight anyway, hoping it'll be the real thing (overnight dialysis).
Khalid has been gone on a trip the last few days. He comes back tomorrow. The other news is that I got to eat out at a Chinese restaurant with labmates: Yuan, Yvonne, Han and Victor. I got to eat pork dumplings! My favorite. I also got egg fried rice. It was so good, and I felt so nice and full afterwards. They also had tea, which was delicious. Best lunch I've had all year.
